In its phosphorylated form, IDO1 acts as a signaling molecule in a pathway that induces long-lasting immunosuppressive effects via genomic mechanisms in plasmacytoid dendritic cells (pDCs). Whether the two activities, i.e., catalytic and signaling, are spatially segregated has been unclear. We found that, under conditions favoring signaling rather than enzymic activity, IDO1 switches from cytosol to early endosomes.
This event required direct interaction with and activation of class IA phosphoinositide 3-kinases (PI3Ks), which contributed to the full expression in vivo of the immunoregulatory phenotype conferred on pDCs by the IDO1 signaling pathway.
Thus IDO1’s spatial dynamics, besides accommodating the needs for short- vs long-lasting mechanisms controlling acute vs chronic inflammation, respectively, may open a completely new perspective in IDO1-based therapies.
